In vitro generation of human cytomegalovirus pp65 antigenemia, viremia, and leukoDNAemia.
Immunocompromised patients with disseminated human cytomegalovirus (HCMV) infection have circulating PMN carrying HCMV pp65 (antigenemia), infectious virus (viremia), and viral DNA (leukoDNAemia). Because HCMV does not fully replicate in PMN, it is generally hypothesized that virions and viral materials are taken up by phagocytosis from fully permissive HCMV-infected endothelial cells. However,...
متن کاملEvaluation of an abbreviated protocol for cytomegalovirus pp65 antigenemia testing.
The cytomegalovirus antigenemia assay remains a useful tool for monitoring reactivation among transplant recipients. In this study, we compared protocols using direct lysis of small-volume, whole blood (WB) samples vs peripheral blood leukocyte (PBL)-enriched fractions. Of 363 evaluable samples, 51 (14.0%) were positive by one or both methods. Sensitivity, specificity, and negative and positive...
متن کاملStandardization of the human cytomegalovirus antigenemia assay by means of in vitro-generated pp65-positive peripheral blood polymorphonuclear leukocytes.
We generated in vitro human cytomegalovirus (HCMV) pp65-positive polymorphonuclear leukocytes (PMN) resembling those detected in vivo, following cocultivation of PMN from healthy donors and wild-type HCMV-infected endothelial cells or fibroblasts. After purification, PMN are suitable for preparation of cytospots which can be used for the antigenemia assay. Cytospin preparations containing a pre...
متن کامل2-Hour cytomegalovirus pp65 antigenemia assay for rapid quantitation of cytomegalovirus in blood samples.
Of 109 blood samples tested for cytomegalovirus (CMV) antigenemia, 18 (16.5%) were positive. CMV Brite detected 13 and CMV Brite Turbo detected 16 of the 18 positives. There was no significant difference in the number of positive cells detected per sample. The seven discrepant samples contained a median of only one positive cell.
متن کاملComparison of the performance of polymerase chain reaction and pp65 antigenemia for the detection of human cytomegalovirus in immunosuppressed patients.
INTRODUCTION Human cytomegalovirus (HCMV) is often reactive in latently infected immunosuppressed patients. Accordingly, HCMV remains one of the most common infections following solid organ and hemopoietic stem cell transplantations, resulting in significant morbidity, graft loss and occasional mortality. The early diagnosis of HCMV disease is important in immunosuppressed patients, since in th...
